We recently published a new inducible plasmid-based transgene expression system that includes a Tet-on-Tet operator under control of a minimal promoter (Mta). Here we describe a novel transgene expression system that includes a Tet-on-Tet operator and a TATA-TATA box. This system is used to study the effect of doxycycline treatment on the expression of the TATA-TATA box in human and mouse embryonic cells. In these cells, a tetracycline responsive promoter was found and this system was used to study the effect of doxycycline treatment on the expression of Tet-on-Tet. The system also showed a low background level of doxycycline-induced gene activation.
